摘要
目的:探讨炎症性肠病(IBD)大鼠模型肠黏膜TRPV1表达和肥大细胞变化,以及二者在IBD发病机制中的作用。方法:雄性SD大鼠用Morris法制备炎症性肠病模型,免疫组化法检测大鼠结肠黏膜TRPV1蛋白表达,RT-PCR检测TRPV1 mRNA表达,并设正常对照组。结果:正常组肠黏膜无或弱阳性TRPV1表达,IBD组肠黏膜TRPV1 mRNA和蛋白表达较正常组增强,肥大细胞数显著增多(P<0.01)。TRPV1蛋白及mRNA水平与肥大细胞数量之间呈正相关(r1=0.528,r2=0.62,P<0.01)。结论:IBD发生发展与TRPV1过度表达及肥大细胞数量变化密切相关。
Objective To investigate the expression of TRPV1 in colonic mucosa of rats with inflammatory bowel disease(IBD) and the changes of mast cells,and their roles in the pathogenesis of IBD.Methods The IBD model were established with Morris method in male SD rats.The expression of TRPV1 mRNA and protein in colonic mucosa were examined by Semi-quantitative RT-PCR and immunohistochemistry,respectively.The control group was set.Results Compared with normal group,TRPV1 mRNA and protein of IBD group was overexpressed,the mast cells quantity in IBD group was significantly increased(P<0.01).The expression levels of TRPV1 protein,mRNA and the quantity of mast cells was positively correlated(r1=0.528,r2=0.62,P<0.01).Conclusion The pathogenesis of IBD were closely related with the overexpression of TRPV1 and the quantity of mast cells.
