摘要
通过测定谷丙转氨酶(GPT)活力的方法,观察了牛磺熊去氧胆酸(TUDCA)对GPT的作用。发现TUDCA对GPT呈抑制作用。且随其浓度的增加,抑制作用增强,当TUDCA浓度为0.833mg/ml时,对GPT的抑制率达到81%。动力学实验还表明,增加底物浓度,并不能消除TUDCA对GPT活力的抑制作用,K_m值不变,V_m值作减小,K_m/V_m值增大,呈现出非竞争性抑制作用。同时用CCl_4小鼠肝损伤模型,观察了口服TUDCA与UDCA对皿清GPT活性的影响,提示TUDCA对肝脏无损害,较UDCA为优。
This paper deals mainly with the effects of TUDCA on activities of GPT.The studies show that TUDCA is a kind of compounds which inhibits activities of GPT.The rate of innibition is 81% at 0.833 mg/ml concentration of TUDCA.In the kineties studies,we found it impossible to relieve the inhibition depending on increasing the concentration of substrate.The value of Km/Vm in TUDCA-bearing reacion is higher that in normal.Further more,we studied the activites of TUDCA in mice.The results show that TUDCA is harmless to liver.
