摘要
脊髓内神经降压素(NT)在痛觉调制过程中发挥重要作用。(1)大鼠脊髓蛛网膜下腔注射NT,可显著增强电针镇痛效应,用ANTS预处理动物40min后,电针镇痛作用明显低于对照组。(2)脊髓蛛网膜下腔注射吗啡10μg,NT的电针镇痛作用明显加强,而用100μg纳洛酮预处理后的大鼠,电针10min注射NT,大鼠痛阈降低非常显著(P<0.01)。(3)电针大鼠双侧"足三里"10min,向脊髓蛛网膜下腔注射DynA(3ug)和NT(2ug),注药后30min电针期间,大鼠痛阈明显升高(P<0.01),用ADgnAS中和脊髓释出的DynA,明显降低了NT增强电针镇痛的作用。结果表明:脊髓内NT参与电针镇痛,脊髓蛛网膜下腔注射NT增强电针镇痛的作用和内源性阿片肽有关,而上述NT在电针镇痛中作用,在脊髓内至少一部分作用可能是由强啡肽A中介的。
Neurotensin (NT) plays an important role in the analgesia in the spinal cord. The effects of intrathecal injection of NT, antineurotensin seruxn(ANTS),Morphinn(M),Napoxone(Nx),Dynorphin A1-13(Dyn A),and Anti- Dynorphin A serum (ADyn AS) on the pain threshold(PT) and EA analgesia in rats were investigated in this paper.The potassium iontophoresis induced tail -flick was used to measure the pain threshold.① When NT was intrathecally injected during EA analgesia, the pain threshold was dramatically increased of 142.70%~183.97 %(P<0.01).After injection of ANTS, EA analgesia only produced an increase of 9.41%~25.88 % in pain threshold, which was decreased significantly as compared with that of NRS-EA group. ② Morphin strengthens EA analgesia induced by intrarhecal injection of NT.And that NT enhanced the EA analgesic effect was antagonized by injection of Nx (100 ug).③ While injecting Dyn A (3 ug) plus NT(2 ug)during EA analgesia, the pain threshold was remarkably increase of 146 .17%~223.79%(p<0.01)as compared withthat of control groups. The pain threshold in ADyn AS-NT-EA group was only increased of 39.52%~90 .75 % , which was became weakened significantly as compared with that of NRS-NT-EA group.Our results indicated that intrathecal injection of NT enhanced theEA analgesia effect, and the EA analgesia effect induced by injectionof NT may be mediated, at least partly, by Dyn A in spinal cord of the rat.
