摘要
报道测定血浆中常咯啉的反相离子对高效液相色谱法。用十八烷基键合相柱,以0.01 mol/LHClO_4水溶液-甲醇(45:55)作流动相(pH 2.30),以常咯啉合成时的中间体为内标。血浆浓度检测限可达25ng,重现性较好,空白和给药兔血浆的回收率分别为98.80±5.99%和95.46±7.23%,cv=4.68±3.16%和4.70±2.62%。人空白血浆的回收率为87.10±6.49%,cv=3.97±2.28%。
A reversed phase ion-pair HPLC determination of changroline in
plasma with ODS column was described. Perchloric acid was used as the coun-
ter ion and 0.01 mol/L perchloric acid aqueous solution-methanol(45.55, pH 2.30)
as mobile phase. 4-(4'-Hydroxyanilino)-quinazoline was chosen as internal stand-
ard. Detectability of plasma sample was 25 ng. The specificity and sensitivity
of the method were rather good, from the plasma samples of rabbits spiked with
changroline and those of the changroline administration the recovery was 98.80
±5.99% and 95.46±7.23%, the coefficient of variation was 4.68±3.16%
and 4.70±2.26%, respectively, as well the recovery from plasma of men was
87.10±6.49%,the coefficient of variation was 3.97±2.28%.The method was
useful for preliminary pharmacokinetic and clinical studies.
