摘要
本实验用氢过氧化枯烯作用于培养的人脐带静脉内皮细胞,探讨脂质过氧化对血管内皮细胞的作用。结果表明,氢过氧化枯烯激发和促进了内皮细胞的脂质过氧化过程,使内皮细胞的过氧化脂质含量升高。在形态结构上内皮细胞出现收缩和生物膜系统的损伤。用放射免疫方法测定,内皮细胞培养液中的6-keto-PGF_1α含量减少,表明PGI_2的产生受到抑制。上述变化可认为:内皮细胞对脂质过氧化的作用是敏感的,提示脂质过氧化通过损伤内皮细胞,可能在动脉粥样硬化发病机理中起重要作用。
The effect of cumene hydroperoxide on cultured endothelial cells (ECs) from human umbilical vein was investigated both morphologically and functionally. The results showed that cumene hydroperoxide stimulated and faciliated the lipid peroxidation in ECs. The concentration of lipidperoxide in ECs increased with the increase of cumene hydroperoxide. At the same time, the ultrastructural changes were contraction, plasma membrane blebbing, dilatation of endoplasmic reticulum and mitochondria, mainly leing the changes of biological membrane. The concentration of 6-keto-PGF_1α in the medium increased slightly in lower concentration group of cumene hydroperoxide (0.1mM), whereas it decreased remarkably in the higher concentration group. It illustrated that the biosynthesis of prostacyclin was inhibited.
