摘要
用RP-HPLC法分离并测定了牛蒡子中5种2,3-二苄基丁内酯型木脂素——牛蒡甙(Ⅰ)、牛蒡酚A(Ⅱ)、牛蒡酚F(Ⅲ)、牛蒡甙元(Ⅳ)、牛蒡素B(Ⅴ)。以安定为内标,分析柱C18,甲醇-水-乙腈-四氢呋喃(57:49:11:1)为流动相,梯度流速,1.0~1.5m1/min。检测波长220nm,线性范围0.029~0.242μg,相关系数r=0.9993~0.9999,回收率96.14~104.99%。本法简便、快速、灵敏。
Arctium lappaol Linne is a
kind of traditional Chinese medicine used as anticancerand
antinephritis agent.Twelve lignans have been isolated from its seeds.
A reversed phase HPLC method for the separation and determination of
the five lignans:arctiin(Ⅰ) ,lappaol A(Ⅱ),lappaol
F(Ⅲ),arctigenin(Ⅳ),neoare B(Ⅴ)in the seeds of Arctium lappaolLinne
is described,The column was 25 cm × 6 mm ID, packed with ODS(10μm).
The columntemperature was 28℃. The mobile phase was a mixture of
methanol-water-acetonitrile-tetrahydrofuran(57 :49 :11 :1).The flow
rate was 1.0~1.5 min-1. Diazepam was used asan internal standard,UV
detection was performed at 220 nm. The relative standard deviation of
the method was 2.16~2.96%. The calibration curves showedgood
linearity over the range of 0.029~0.242μg,r=0.9993~0.9999 and the
detection limitswere 3.38~4.20 ng.The recoveries were 96.14~104.
99%for the five lignans. The method is simple,fast, sensitive and
accurate and has been applied to the analysis of Arctiumlappaol Linne
of different sources.It can also be used for the quality control of
Arctium preparations forclinical uses.
