摘要
iv甲基莲心碱(Nef)6 mg/kg于正常血压、肾性高血压、DOCA盐型高血压大鼠,在血压下降的同时,伴有LVSP,±(dp/dt)max,(dp/dt)P^(-1)的降低,正常血压及肾性高血压大鼠LVSP^-(dp/dt)P^(-1)环缩小,其斜率变小,心率短时变慢,LVEDP变化不明显。iv Nef 2 mg/kg于Wistar高血压大鼠时,仅明显降低血压,而对其它血流动力学指标无明显影响,Nef降低舒张压的作用大于降低收缩压。猫后肢灌流实验证明Nef有明显降低血管阻力的作用。结果表明Nef的降压作用可能由于扩张外周血管所致。
Neferine is an alkaloid isolated from the green seed embryo of Nelumbo nucifera Gaertn.Neferine possesses a hypotensive effect in experimental animals.The present study was designed to investigate the influer.ce of neferine on the left ventricular function and peripheral bloodvessel resistance in rats.Neferine (6mg/kg)given intravenously to pentobar'bital anesthetized normotensive rats,DOCA-salt and renal hypertensive rats decreased blood pressure,LVSP,±(dp/dt)max; diminished LVSP-(dp/dt)/P-1 loop and heart rate,while LVEDP was not changed significantly. Neferine (2 mg/kg)given intravenously to rats decreased only blood pressure markedly.At the dose of 0.6 mg/kg neferine produced an obvious reduction of the preipheral re-sistence in the perfused hind legs of cats.In addition,the extent of decreasing diastolic blood pressure was greater than that of systo-lic blood pressure.The results suggest that the drug inhibits the cardiac contractibility in high dosage,and it reduces the peripheral vessels resistance
