糙皮侧耳和佛罗里达侧耳菌丝原生质体的形成与再生 被引量:1

FORMATION AND REGENERATION OF PROTOPLASTS OF PLEUROTUS OSTREATUS AND PLEUROTUS FLORIDA

摘要 本文报道了使用蜗牛酶和纤维素酶从糙皮侧耳和佛罗里达侧耳双核菌丝体中,成功地分离了高产量的原生质体。菌丝体培养时间,混合酶液 pH 值,一定浓度的酶液用量,酶解温度和时间与原生质体形成相关。原生质体的两种再生型被观察到,并计算了再生频率,糙皮侧耳为2.5%.佛罗里达侧耳为2.3%。 The formation and regeneration of mycelial protoplasts from Pleu- rotus ostreatus and P.florida using snailase and cellulase were studied.The amount of release of protoplasts is related to mycelial age,buffer pH,temperature and dose of enzyme used.To achieve a better enzymolysis result,young mycelia, botained from the edge of colonies which were incubated on solid plates at 25℃ for 4 days,were used in this study.5ml 1% of snailase and cellulase was added to one gram mycelia (wet weight) in pH5.8 buffer for enzymolysis at 35℃ for 4.5 hours.The regeneration frequeucy of protoplasts of both strains were observed by using RM medium,2.5% for Pleurotus ostreatus and 2.3% for P.florida. Two regeneration types were obtained in LRM medium.For the first type,proto- plasts directly produced one or more germ tubes; but for the second,protoplasts produced bud-like structures at beginning and then develo ped into germ tubes.
