摘要
本文介绍了一种新的生物材料血液相容性体外评价方法,用特异抗人血小板膜糖蛋白(GPⅢa)的单克隆抗体SZ-21以固相放射免疫法对人血清白蛋白、人血纤维蛋白原及鼠尾胶原镀膜材料管表面粘附血小板进行定量测定。 试验结果说明本研究建立的新方法可快速、准确、定量地测定生物材料吸附血小板的性能,并为进一步深入研究生物材料对蛋白抗原决定簇的影响提供了新的手段。
<ABSTRACT>
A new method is described to quantitate platelets adhering on biomate-
rial surfaces in vitro, based on a surface phase radioimmunoassay using
monoclonal antibody (McAb ) SZ-21 to human platelet membrane glycopro-
tein complex (GP Ⅲa ).
Results showed that after 5 min perfusion at 2,000S-1, platelet deposi-
tion determined using anti-human platelet antibody (125I-SZ-21 ) is (plate-
let/mm2 ):4, 173±932 (A ); 59, 032±125, 554 (Fbg ): and 71, 252±11, 484
(Col ). These results correlate well with those obtained using 51Cr labeled
platelets (r = 0. 98 ), and morphometric evaluation using SEM.
Using a straight linear correlation between deposited platelets (51Cr ) and
immunoadsorbed antibody (125I-SZ-21 ), platelets adhered on surfaces of 4
polymers were determined (platelet/mm2 ): 19, 493±2,050 (Sil); 48, 193±4,
055 (PTFE ); 50, 375±8, 675 (PYC ); and 101, 906±5, 916 (PE ).
The new method has advantages of simplicity for measurcment of plate-
let deposition in whole blood, since platelet separation, washing and label-
ing procedures are eliminated, thus ensuring possible selection of platelet
populations and modification of platelet functions.
