摘要
用CT-HRP逆行追踪法及ChAT单克隆抗体的ABC免疫组织化学技术研究了新生大鼠骨骼肌提取液(ME,20~50kD)对钳夹大鼠坐骨神经所致的腰骶部脊髓腹角和背根节神经细胞溃变的影响。实验结果显示:注射ME实验组的脊髓腹角运动神经元和背根节细胞的存活均数与注射盐水的对照组相比,其比值为6:0,有显著性差异(P<0.01)。提示骨骼肌提取液(20~50kD)对脊髓腹角运动神经元和背根节感觉神经细胞的演变有明显的保护作用。
The effect of a skeletal muscle extract (20~50kD) on the degeneration of lumbar spinal motoneurons and dorsal root gan glion cells after sciatic nerve crush was studied by using CT-HRP retrograde tracing method and a specific choline acetyltransferase (ChAT) monoclonal antiserum immunoreactive ABC method. The skeletal muscle extract (ME) was prepared from limbs of 80 rat newborns. The homogenate was centrifuged and filtrated, theT, a chromatography and electrophoresis was made to determine the molecular size. Each fraction of 10~50 kD was tested on spinal cord cell cultures from E12 rat embryos. Eighteen rats were divide into 3 groups. The sciatic nerve was crushed with hernoctatic clamps and ME was injected under the epineurium in the rats of group A and the ipsilateral of group B, the sodium solution was injected in group C and the contralateral side of group B as control. Following a postoperative period of 4 weeks , the result showed that the average number of survival motoneurons in the lurnbar spinal cord of rats in ME injected group A was evidently higher than that of control (12.8: 7.7), which is statistically singnificant P<0.001. The tnotoneurons were full of TMB crystalline aggregates and POsitive ChAT irnmunoreactivity, the a-cons and dendrites were much thicker and longer than that in control. The average number of survival lumbar DRG neurons (including tile large A kind cells and small B kind cells) in the rats of ME experimental group A was 32, and that in control group was 12, which is statistically singnificant P<0.01. It is inferre<l from the result that the neurotrophic factors in the skeletal muscle extract (20~50 kD) could improve the regeneration for injured spinal cord rnotoneurons and dorsal root ganglion neurons.
