微波固定对琥珀酸脱氢酶细胞化学影响的电镜观察 被引量:2

Electron microscopic observation of cytochemical effect of microwave fixing on succinodehydrogenase

摘要 对浸泡在四种不同液体(生理盐水、0.25%戊二醛、2.5%戊二醛、5%戊二醛)中的鼠肝组织进行微波照射固定,并按琥珀酸脱氢酶(SDH)常规孵育、制片、电镜观察。结果表明:四个组都随微波照射时间增加,液体温度不断升高,肝细胞内线粒体SDH酶活性由强逐渐减弱,定位趋于稳定;其中生理盐水SDH酶活性保存最好,且以微波照射40s时,SDH酶活性及定位最佳,超微结构清晰。 Liver tissues of rat in four different kinds of liquid(normal saline,0. 25% GLA,2.5%GLA,5%GLA)were fixed by mesns of microwave irradiation. Incubation, process and observation were made according to succinodehydrogenase(SDH)routine. The results indicated that the four groups shared the same rule that with the increase of irradiation time and the liquid temperature raised continuallg, the activity of SDH in mitochondrion of liver cells decreased,but the localization become stabilized.The activity of SDH was best preserved in normal saline group, better than any other GLA group, and when irradiation time was 40 s,in the same group we got the best activity and localization of SDH,and the ultrastructure was very clear.
