摘要
采用全细胞膜片钳技术研究颅痛定对酶解分离的豚鼠心室肌细胞膜钙通道电流(I_(Ca))的影响.结果:当保持电位为-40mV、去极化从-3O至+60mV、刺激脉宽为150ms、频率为1Hz时可记录到I_(Ca).维拉帕米10μmol/L可明显抑制I_(Ca).颅痛定1~100μmol/L可浓度依赖性抑制I_(Ca),10、30μmol/L分别使I_(Ca)峰值从-1135±224降至-832±216pA(n=7,P<0.01),从-1174±216降至-452±71PA(n=8,P<0.01),抑制百分率分别为27%、61%,冲洗后可部分恢复.提示颅痛定对I_(Ca)的抑制为其主要的抗心律失常作用机理之一.
Effects of 1-tetrahydropalmatine (1-THP) on calcium channel currents (Ica) were studied in the enzymatically dispersed guinea pig ventricular myocytes using the whole-cell patch-clamp method. When the holding potential was -40mV, cells were depolarized to + 60mV for 150 ms at frequency of 1 Hz,slow inward currents were observed. Verapamil 10 μmol ?L-1 inhibited significantly Ica. 1-THP l-100μmol ?L-1 inhibited Ica in concentration-dependent manner. 1-THP 10,30μmol ?L-1 decreased Ica from -1135±224 to-832±216 pA (n = 7,P<0. 01),and from-1174±216 to-452±71 pA(n = 8,P<0. 01) ,respectively, Ica partially increased again after washout of 1-THP with tyrode solution. The results suggested that 1-THP was a calcium antagonist from natural resource.
