摘要
本文报告人低分化鼻咽癌裸小鼠移植模型的建立及其生物学特性的研究,实验动物为NC系裸小鼠(少量Swiss系),使用CNE—2 Z细胞(1×10~6/0.5ml细胞)皮下接种,历时二年,在203只动物传代30次,移植成功率为100%。组织学及超微结构显示为低分化鳞状细胞癌特征,染色体显示人类肿瘤细胞染色体,有EB病毒基因组存在及EB病毒核抗元(EBNA)、膜蛋白(LMP)表达。我们所建立人低分化鳞状细胞癌裸鼠移植模型,具有移植成功率高,呈进行性生长,恶性程度高,不会自行消退等特点,可供进一步研究鼻咽癌生物学特性及实验性治疗之用。
The establishment and characteristics of a transplantable model of human epithelial cell strain of poorly differentiated nasopharyngeal carcinoma(CNE-2Z) in nude mice was reported. The experiments were made on 1-3 month-old nude mice (NC and Swiss) of both sexes. The CNE-2Z cells (1?10 6 /0.5ml) were inoculated subcutan'eously . The transplanted tumor had been passed up to 30 generations in 203 recipient animals and subsequent serial transplantability was 100%, despite of origin, age of nude mice and inoculated route ( s.c.,i.P, i. m,).The latent period was shortened from 20 to 5- 14days. Then the slow, rapid and late growth periods were 2 - 3,4 - 8 and 9-10 weeks, respectively. The longest survival period of tumor-bearing mice was 144days and the greatest weight of graft was 19.7g, which was two thirds the weight of the host. Tumor tissue was easy to infiltrate to neighbouring tissues, and to metastasize into lymph nodes and lungs. The graft had characteristics of p oorly differentiated squamous carcinoma on the light and electronic microscopy. The chromosome showe. ed a karyotype of human tumor. There are existence of Epstein-Barr virus genes and expression of EBV-encoded protein (EBNA 1& LMP) in the transplanted tumor. The model can be used for the further study human nasopharyngeal carcinoma.
