摘要
目的:探讨蒺藜皂苷(简称STT)对体外培养的肾癌细胞(786-O)的影响及其抑癌机制。方法:应用MTT法、Wright染色及AO荧光染色法和流式细胞术(FCM)及结合免疫荧光分别测定STT对细胞的生长抑制作用,细胞凋亡的形态变化,细胞周期及Bcl-2蛋白表达的影响。结果:MTT法显示STT可对786-O产生明显的细胞毒作用;Wright染色及吖啶橙荧光染色法发现STT可诱导786-O在体外产生凋亡;STT可使786-O的S期细胞比例增多;STT能够降低786-O细胞的抗凋亡蛋白Bcl-2及细胞周期蛋白D1的表达。结论:STT对786-O细胞有明显的体外生长抑制作用,并通过影响细胞周期及下调Bcl-2蛋白表达而诱导细胞凋亡。
Objective: To investigate the effect of saponins from Tribulus terrestris (STT) on the renal carcinoma ce11(786.O) in vitro, and inhibitory mechanisms. Method: Effects of STT on the cytotoxicity, morphological changes of apoptosis, cell cycle and expression of Bcl-2 protein in the 786-O were tested respectively by MTT method, Wright and acridine orange stain assay, as well as flow cytometry(FCM). Resuit: After the 786-O was treated by STT, it was shown that: 1) A significant cytotoxic effect was observed by MTT assay; 2) Apoptosis-induced was viewed by Wright and acridine orange stain assay; 3) The distribution of 786-O on S phase was increased: 4) The expression of Bcl-2 protein and cyclin DI was decreased, conclusion: STF can significantly inhibit the growth of 786-O in vitro, partially, by apoptosis.
