摘要
目的:探讨参麦注射液(SM)对大鼠心肌缺血-再灌注损伤的防治作用及机制。方法:结扎冠状动脉前降支复制大鼠心肌缺血-再灌注损伤模型,描记标准肢体II导联,测定心肌组织中丙二醛含量及结构型、诱导型一氧化氮合酶(cNOS,iNOS)活性,逆转录聚合酶链反应(RT-PCR)法测其cNOSmRNA、iNOSmRNA表达。结果:SM组心律失常发生率明显低于其他各组;cNOS活性及其mRNA表达显著增高(P<0.01),iNOS活性及其mRNA表达显著降低(P<0.01)。结论:SM能减轻缺血-再灌注引发的损伤,其作用机制可能与增加cNOS活性,促进cNOSmRNA表达,抑制iNOS活性,降低iNOSmRNA表达有关。
Objective: To study the protection of Shenma injection against myocardial ischemia-reperfusion injury on rats and its mechanism. Method: The left anterior coronary artery was ligated to establish the animal model of myocardial ischemia-reperfusion injury. During the process, the electrocardiogram was traced. Also the content of malondiadehyde(MDA), activity of constitutive nitric oxide synthase (cNOS) and induced nitric oxide synthase (iNOS) in myocardium were examined. The expressions of cNOS mRNA and iNOS mRNA in myocardium were detemined by the reverse transcription-polymerase chain reaction(RT-PCR). Results: Shenma injection obviously decreased the oceurence of arrhythmia, increased the activity of cNOS, the expression of cNOS mRNA( P〈 0.01 )and inhibited the activity of iNOS and reducing the expression of iNOS mRNA(P〈0.01). Conclusion: Shenma injection has protective effect on myocardial ischemia - reperfusion injury in rats. The mechanism may be related to increasing the activity of cNOS, promoting the expression of cNOS mRNA and inhibiting the activity of iNOS and reducing the expression of iNOS mRNA in ischemia-reperfusion injury myocardiurn.
