摘要 取不同基因型的东北主栽玉米自交系胚叶3mm接种在诱导培养基上,研究东北主载的玉米胚叶再生体系的建立。结果表明,玉米胚叶愈伤组织的形成能力因其形态学部位的不同而呈显著差异,从基部向上依次递减;愈伤组织的形成还与基因型和植物激素配比有关,其中,2,4-D是最重要的因素,加入一定浓度的AgNO3有利于愈伤组织的诱导。探讨了影响不定芽分化的因素,初步建立了玉米胚叶的再生体系。 3 mm long leaf segments of Zea mays L. from 7-day-old seedlings were cultured on inducing nutrient medium to study on establishment of regeneration system of maize embryo leaf of inbred lines in northeast. The results showed that the ability of callus induced maize embryo leaf was obviously different from positions, genotype and plant hormones at different concentrations. Inducing rate of embryogenic callus gradually reduced from bottom to top of embryo leaf. 2, 4-D was the most factor to callus induction. In addiction, the medium supplemented with certain AgNO3 was profitable to induce callus tissues. Now we discussed factors of affecting differentiation of shoots and established regeneration system of maize embryo leaf of inbred lines in northeast.
