摘要
反义RNA技术是一种有效的遗传操纵手段,可用于鉴别克隆基因的功能并培养新型的植物品种。通过DNA体外重组的方法,把拟南芥植物中分离出的腈水解酶、泛素转移蛋白,环菲林、亮氨酸氨肽酶反义基因克隆到双元载体pBI121中,构建了含上述4种反义基因的嵌合载体用于遗传转化。限制性内切酶酶谱分析和DNA序列分析证实了这些新构建的嵌合载体含反义基因。通过土壤农杆菌介导法获得了泛素转移蛋白,环菲林,亮氨酸氨肽酶反义基因的转基因拟南芥植株。
The antisense RNA technique can be efficiently used to identify previously un known genes and produce new types of plant by down-regulation of endogenous plant genes. In or der to study the function of nitrilase, ubiquitin carrier protein, leucine aminopeptidase and cyclophilin genes in Arabidopsis plants, the chimeric vectors of these antisense clones have been constructed and introduced into Arabidopsis via Agrobacteriurm tumefaciens-mediated transformation.These structures were confirmed by restriction mapping and DNA sequencing. Some transgenic plants have been produced.
