摘要
目的 建立家兔血浆中酮康唑(ketoconazole)浓度的高效液相色谱检测法。方法 采用HypersilODSC18柱,以甲醇 水 三乙胺(79:2 0 :1)为流动相,紫外检测波长2 4 1nm ,安定为内标,乙醚提取,浓缩后进样。结果 酮康唑与内标分离良好;血浆酮康唑浓度在0 .5~10 0 .0 μg/ml范围内线性良好,r =0 .9996 ;最低检测浓度为0 .2μg/ml;0 .5、5 .0、5 0 .0 μg/ml三种浓度的平均回收率分别为91.2 %、96 .9%和98.7% (n =5 ) ;日内和日间RSD <10 %。结论 该方法操作简便、可靠、取血量少,适用于酮康唑的血药浓度测定和临床生物利用度的研究。
Objective To establish a HPLC method for the determination of ketoconazole level in plasma ofrabbit. Methods An agilent 1100 instrument with Hypersil C18 column(5μm,4.6 mm×250mm) was used.The mobile phase was a mixture of methonal, water and triethylamine in a ratio of 79:20 : 1 (adjusting pH to 6.6 by hydrochloric acid) at a flow rate of 1.0 ml/min. The ultraviolet detector was set at 241nm. Results A good linear range of ketoconazole was obtained from 0.5 to 100μg/ml( r = 0. 9996), and the detection limit was 0.2μg/ml. The average recovery rate( n = 5 )at the concen-tration of 0.5, 5.0 and 50ug/ml was 91.2%,96.9% and 98.7%, respectively. Conclusion The method was simple, accurate and reliable. It is suitable for the determination of ketoconazole level in plasma and can also be used for bioavailability study.
