摘要
临床上肿瘤细胞的抗药性常与编码P-糖蛋白的多药抗性基因有关,为测定肿瘤中多药抗性基因的表达,我们用地高辛标记的cDNA探针通过原位杂交检测了25例肺癌组织中多药抗性基因的表达,发现在12例中多药抗性基因过度表达。结果提示临床上这些肿瘤对化疗不敏感可以用P-糖蛋白介导的药物外流来解释。
ncreased expression of Muitidrug-resistance
(MDR)gene transcripts and of the encoded pro-tein,P-glycoprotein,is found in many types of
tumors,The biological significance of MDR geneoverexpression during the stepwise process of
neoplastic development,however,is ont well under-stood, To assess the possible gignificance
of MDR overexpression in carcinogenesis,we examinedthe cellular distributions of MDR gene
using a cDNA probe by in situ bybridization in 25 lung cancertissues,We found overexpression
of MDR gene was detected in 12 cases. The results of our studyshow that the MDR gene is
expressioned in human lung cancer and suggest that the multidrugtransporter may contribute to
the clinical non-responsiveness of these tumors to chemotherapy.
