摘要
作者运用生化和分子生物学方法,探讨了抗肝炎药联苯双酯(DDB)对人肝癌细胞分化的影响,结果发现:生长在含有10-4mol/LDDB培养液中的人肝癌Bel-7402细胞,其甲胎蛋白(AFP)的分泌量和γ-谷氨酰转肽酶(γ-GT)活力均明显低于培养相同天数的对照细胞,而白蛋白(ALB)的分泌量和酪氨酸-α酮戊二酸转氨酶(TAT)活性又明显高于对照细胞。DDB还能明显增加Bel/7402细胞环化腺苷酸(cAMP)的含量,并抑制癌基因c-myc及肝癌标志AFP基因的表达,增强了P53抑癌基因的表达。上述结果提示DDB能诱导人Bel-7402肝癌细胞的分化。
When human Bel-7402 hepatocarcinoma cell line grew in a medium containing 10-4M DDB,the secretion of α-fetoprotein(AFP) and the activity of γ-glutamyltranspeptidase(γ-GT)were significantly lower than the control cells,whereas the albumin(ALB) secretion and the activity of tyrosine-α-ketoglutarate transaminase(TAT)were markedly increased.DDB at the concentration of 10-4M could significantly increase the content of cAMP in Bel-7402 cells,and also suppressed the expressions of oncogene c-myc and hepatocarcinoma marker AFP gene and enhanced the anti-oncogene p53 expression.The results of this paper suggest that DDB has some reversing effects on the phenotypes of human Bel7402 hepatocarcinoma cell line.
