慢性特发性血小板减少性紫癜患者骨髓巨核细胞中c-myc和c-sis的表达 被引量:5

EXPRESSION OF C-MYC AND C-SIS mRNA ON BONEMARROW MEGAKARYOCYTE IN PATIENTS WITH ITP

摘要 应用原位杂交的方法,结合血小板释放产物β-TG和PF_4测定及巨核祖细胞体外培养,对33例慢性ITP患者骨髓巨核细胞中原癌基因c-myc和c-sis的表达及其与β-TG、PF_4和CFU-MK的关系进行了研究。发现,巨核细胞c-sis表达较对照组明显降低,c-myc表达无显著差异。c-sis表达与β-TG和PF_4含量呈负相关。c-myc表达与CFU-MK呈正相关。MK数增高组的CFU-MK正常,c-myc表达增高,而MK数正常组的c-myc、c-sis表达和CFU-MK均降低,提示多数ITP患者骨髓巨核细胞增生,伴有成熟障碍,少数患者巨核细胞的增殖能力降低。 he mRNA expression of c-myc and c-sis onco-genes in bone marrow megakaryorytes (MK) and itscorrelation with β-TG, PF_4 and CFU-MK were assayedin 33 ITP patients by the method of in situ hybridiza-tion. The expression of csis mRNA was depressed inbone marrow MK in ITP patients as cornpared with thatin controls, while there was no difference in the expres-sion of c-myc mRNA between the two groups. The in-crease of &TG and PF. levels showed a negative correla-tion with the expression of c-sis, while the decrease ofCFU-MK number showed a positive correlation with theexpression of cmyc in bone marrow MK. In the groupwith increased number of MK but normal number ofCFU-MK, the expression of c-myc in bone marrow MKwas elevated , while in the group with normal number ofMK, the expression of cmyc and c-sis as well as thenumber of CFU-MK was declined. It suggested that inmost of ITP patients the bone marrow MK were hyper-plasia with a defective maturation , but in a few cases,there was an impairment in the proliferative capacity ofMK.
