干扰素-a对慢性粒细胞白血病细胞作用的体外研究 被引量:2

THE IN VITRO EFFECT OF INTERFERON-a ONCHRONIC MYELOGENOUS LEUKEMIA CELLS

摘要 作者通过体外骨髓半固体和长期液体培养(LTBMC),研究了干扰素-a(IFN-a)对慢性粒细胞白血病(CML)细胞的作用。在半固体琼脂培养体系中,IFN-a能抑制正常对照和CML患者7天和10天CFU-GM,而不抑制14天及其后的CFU-GM。在LTBMC中,于培养开始只加一次IFN-a(10 ̄2U/ml,10 ̄3U/ml,10 ̄4U/ml),对非贴壁层细胞数和基质细胞层的形成均无明显影响,但大剂量IFN-a(10 ̄3U/ml和10 ̄4U/ml组)对非贴壁层的CFU-GM抑制明显;每周连续加用IFN-a(10U/ml,10 ̄2U/ml,10 ̄3U/ml),对非贴壁层细胞数和CFU-GM均有抑制作用,而且IFN-a浓度愈高,贴壁层的形成愈少。上述结果表明:IFN-a优先抑制晚期CFU-GM,对基质细胞也有一定抑制作用。 he in vitro effect of interferon-a(IFNa) on chronicmyelogenous leukemia (CML) were assayed in semi-solid and longterm bone marrow culture (LTBMC).In semi-solid agar culture systems IFN-a showed in-hibitory effects on day 7 and day 10 CFU-GM growthof normal and CML bone marrow , but little or no ef-fects on day 14 or later CFU-GM growth. In LTBMC ,there showed no inhibition on the cell number of non-adherent layer and the formation of adherent layer ifIFN-a was added only once at the initiation of the cul-ture, but the large dosage of IFN-a (10 ̄3U/ml and10 ̄4U/ml) showed significant inhibitory effects onCFU-GM growth of non-adherent layer. The inhibi-tion on the cell number of nona-dherent layer and theformation of adherent layer was observed when IFN-a(10U/ml,10 ̄2U/ml , 10 ̄3U/ml) was added weekly , andthe higher concentration of IFN-a was added , the few-er of the adherent layer was formed. It suggested thatIFN-a selectively inhibited the late CFU-GM and thedevelopment of stromal cells .
