摘要
应用枯否氏细胞、肝细胞共同培养Rh-123荧光图像分析技术研究急性胆管炎早期枯否氏细胞反应对肝细胞能量代谢的影响。结果表明:大鼠急性胆管炎早期激活的枯否氏细胞对正常肝细胞线粒体能量代谢有明显抑制作用,TNF单抗能在一定程度上减轻这种抑制效应,但不能使肝细胞线粒体能量代谢功能完全维持正常。表明急性胆管炎早期肝脏能量代谢障碍与枯否氏细胞的激活有关,TNF的过度分泌可能是其中的机制之一。
Influence of Kupffer cell(KC )
activationon mitochondrial function of hepatocyte(HC)in acute cholangitis was studied by KC-HC
co-culture and fluorescence image analysis tech- nique.Activated Kupffer cells isolated fromthe
rats with cholangitis for 12 h demonstrat- ed an obvious inhibitory effect on the
normalhepatocyte mitochondria energy metabolism. AntTNF monoclonal antibody
amelioratedthe inhibition but could not maintain hepato-cyte mitochondrial function in a
completelynormal state. The result suggests that activat-ed Kupffer cells play an important role
in thedepression of hepatocyte mitochondrial func-tion during acute cholangitis and
uncheckedTNF production may account for its mecha-nisms.
