摘要
作者采用新生纯系小鼠颅骨体外培养模型,综合利用钙转换率和组织形态学观察等指标,客观地评价了白细胞介素-1(IL-1)调节骨吸收的作用。结果表明,IL-1具有强烈的刺激骨吸收的活性,其两种不同形式IL-1α和IL-1β的活性接近。200~4000ng/ml的白细胞介素-1受体拮抗剂(IL-lra)能完全阻断20ng/mlIL-1α刺激骨吸收的作用,即使是在加入IL-1α4小时后,再加入IL-lra同样能阻断IL-1α诱导的骨吸收作用。此外发现,IL-1在其作用最初24小时内,未能增加培养颅骨骨吸收活性;培养72小时后组织学观察颅骨破骨细胞数量增多。揭示IL-1刺激骨吸收的作用机制是:刺激破骨细胞前体增殖分化成熟,增加破骨细胞数量,而对成熟破骨细胞缺乏直接作用。
AbstractEffects of interleukin 1(IL-1)on the calciumturnover and histological changes were studied in thein vitro culture system of the neonatal mouse calvari-ae. Both IL-1α and IL-1β were shown to stimulate cal-cium release from the cultured calvaria. 200~4000ng/ml interleukin 1 receptor antagonist(IL-lra)blockedcompletely the effect of 20ng/ml IL-1α on the calciumrelease, Histological examination showed that IL-1 in-creased osteoclastic bone resorption in the culture cal-varia. Both IL-1α and IL-1β were shown to be stimulifor bone resorption with no difference in potency.Itwas of interest to find that IL-1 failed to increase boneresorption during its first 24 hours of incubation.These would suggest that IL-1 stimulates bone resorp-tion by increasing osteoclast number rather than theactivity of preexisting mature osteoclasts.
