红景天甙对乙醛刺激的大鼠肝星状细胞激活后功能的影响 被引量:5

摘要 目的:观察红景天甙对乙醛刺激的大鼠肝星状细胞(hepaticstellatecell,HSC)增殖、细胞周期、Ⅰ型胶原蛋白分泌及TGFβ1mRNA表达的影响.方法:常规细胞培养,红景天甙对乙醛刺激的HSC进行处理;以MTT比色法检测细胞增殖变化,流式细胞仪检测细胞周期,ELISA法检测HSC内Ⅰ型胶原蛋白的分泌,RTPCR方法检测HSC内TGFβ1mRNA表达.结果:红景天甙可明显抑制乙醛刺激的HSC增殖;抑制乙醛刺激的HSC由G1→S期转化;抑制HSC内Ⅰ型胶原蛋白分泌和TGFβ1mRNA表达.结论:红景天甙抗肝纤维化与抑制HSC增殖,Ⅰ型胶原蛋白分泌和TGFβ1mRNA表达有关. AIM: To investigate the effect of salidroside, a traditional Chinese medicine, on the cell cycle, cell proliferation, type Ⅰ collagen's secretion and TGFβ1 mRNA expression of rat hepatic stellate cells (HSCs) stimulated by acetaldehyde. METHODS: HSCs stimulated by acetaldehyde were treated with salidroside. The cell cycle was analyzed by flow cytometry, cell proliferation was assessed by MTT colorimetric assay, type Ⅰ collagen of culture medium was detected by enzyme-linked immunoadsordent assay (ELISA) and TGFβ1 mRNA expression was examined by RT-PCR. RESULTS: Salidroside (1.5 g/L) significantly inhibited the proliferation and provoked G0/G1 phase arrest of HSCs stimulated by acetaldehyde and markedly inhibited type Ⅰ collagen's secretion and TGFβ1 mRNA expression of HSCs stimulated by acetaldehyde. CONCLUSION: Salidroside inhibits the proliferation of rat HSCs stimulated by acetaldehyde and provokes G0/G1 phase arrest of HSCs. Type Ⅰcollagen's secretion is related to TGFβ1 mRNA expression.
