正交试验法优选猕猴桃果水溶性多糖的提取工艺 被引量:8

摘要 为确定猕猴桃果中水溶性多糖的提取工艺,对提取方法进行了考察,并对料液比、温度、时间等进行了正交实验,通过极差分析,对提取过程中显著影响提取率的因素进行了统计分析.结果表明,超声波预处理30min可以提高糖的提取率,温度和料液比显著影响可溶性多糖的提取率.最佳提取工艺为料液比(W/V)1∶25、温度80℃、时间4h,提取次数为2次.加入体积分数为4%的2mol/L硫酸铜可以很好地去除样液中的杂蛋白,同时糖的损失很少. In order to know the extraction technology of soluble polysaccharides fromChinensis Actinidia.Orthogonal test L9(33)and the analysis of range were applied to analyze the influence of each factor (such as solid-liquid ratio, temperature and time) on the extraction ratio of the polysaccharides. Comparing with classic hot water extraction method, ultrasound assisted extraction method would improve the extracting efficiency; Experimental results indicat- ed that temperature and solid-liquid ratio significantly affected the extraction ratio of the soluble polysaccharides.The optimum solid-liquid ratio was 1∶25, temperature was 80℃and the extraction time is 4 h, the extraction degree was 2 times.4%(V/ V)CuSO4solution (2 mol/ L) could remove the proteins in extraction juice very well while the loss of polysaccharides were quite little.
