摘要
本文建立了反相ＨＰＬＣ测定替硝唑血药浓度的方法，以０．８ｍｏｌ／ＬＨＣｌＯ＿４溶液沉淀蛋白后直接进样，方法简便，平均绝对回收率９５．１％，日间和日内ＲＳＤ％均小于４％，兔静注替硝唑３０ｍｇ／ｋｇ后，体内过程符合二室开放模型，药动学参数分别为α０．７９１６ｈ￣（－１），β０．０６２８ｈ￣（－１），Ｔ＿（１／２）α０．８７６ｈ，Ｔ＿（１／２）β１１．０４ｈ，ＡＵＣ２７２．３μｇ·ｈ／ｍｌ，Ｖｃ０．５６Ｌ／ｋｇ，ＣＬ０．１１Ｌ／（ｈ·ｋｇ）。
Amethod for determination of tinidazole in plasma by high
performance liquid chromatography was estab-lished, using 0.8mol/L HClO_4 as protein
precipitating-agent. The methud was simple. Average recoveries were95.1%with between- days
and within- days lessthan 4%. After 30mg/kg tinidazole was injected in rabbits,
thepharmacokinetic model was described as two compartment model. The pharmacokinetic
parameters were α 0.7916h￣(-1)、β0.0628h￣(-1)、T_(1/2α)0. 876h、T_(1/2β)1 04h、
AUC272.3(μg·h/ml、 V_c 0. 56L/kg、CL0.11L/(h·kg) respec- tively.
