Quantitative Real-time Detection of Spontaneous Interleukin 2 Gene Expression in Peripheral Blood Mononuclear Cell by Polymerase Chain Reaction

摘要 目的:建立白细胞介素2(IL-2)基因表达的绝对定量实时PCR检测技术。方法:应用实时定量PCR方法检测25名健康运动员中IL-2mRNA的表达。主要结果和结论:应用实时定量PCR方法能够对健康受试者外周血白细胞自发性IL-2mRNA表达进行定量分析,相关系数r>0.99。 Objective To develop a method for quantitative real-time detection of interleukin 2(IL-2) using polymerase chain reaction (PCR). Methods Gene expression of IL-2 in peripheral blood mononuclear cell (PBMC) from 25 healthy individuals was detected by PCR. An endogenous “housekeeping' gene GAPDH was used to normalize the results. Results and Conclusion The spontaneous IL-2 mRNA expression in PBMC in a given sample could be measured by PCR with relativity correlation coefficient of r>0.99.
