摘要
目的探讨光动力作用对肿瘤细胞信号转导系统的影响。方法运用放射免疫及酶联免疫的方法,检测光动力作用后H22细胞胞浆蛋白酪氨酸激酶及磷酸酶活性的动态变化,比较各组有无显著差异。结果光动力作用后,H22细胞蛋白酪氨酸激酶活性随时间的延续而出现先升后降的动态变化,而蛋白酪氨酸磷酸酶活性在光动力作用后则相应伴随有一个先降后升的动态变化。结论光动力作用能够促使H22细胞蛋白酪氨酸激酶和蛋白酪氨酸磷酸酶的活性改变,反映了蛋白酪氨酸残基磷酸化与去磷酸化的即时调节机制,提示胞内信号转导系统的激活。
Objective To investigate the changes of signal transduction in tumor cell with photodynamic therapy.Methods Radio-immunity and ELISA were used to dynamically detect the activity of protein tyrosine kinase and phosphatase in the cytoplasm of H22 cells treated by PDT. To know whether there is significant difference in each group.Results PDT inhibited the activity of protein tyrosine phosphatase in H22 cells treated by PDT.The activity of protein tyrosine kinase was increased immediately and returned to the normal level in 120min.Conclusion This study confirms that PDT could change the activity of protein tyrosine kinase and phosphatase. The induction indicates the real-time regulation of phosphorylation and dephosphotylation, it improves the stimulation of signal transduction too.
