摘要
将逆转录病毒载体(pN2-LacZ)或pN2-CMV-ProUK质粒,直接导入Wistar大鼠股四头肌、心肌或动脉管内壁,证明外源性基因可以在上述组织中表达出相应的蛋白质。外源基因在注射入股四头肌后7天表达水平最高,约可持续1~3个月以上。将带有外源基因的医用缝合线,缝合于肌肉组织中,亦可于肌肉组织中表达出相应的蛋白质,其表达效率远高于直接注射法。
N2-LacZ or pN2-CMV-ProUK plasmid DNAwas separately injected into rat
quadricep muscles andβ-galactosidase or Pro-UK was detected after 2 days.The expression of
foreign genes lasted for at least 3months. PN2-LacZ plasmid was injected into the leftventricular
wall of the rat, and the expression of LacZgene was more efficient than in the quadricep muscle.
pN2-LacZ was also introduced into the rat arterialwall by balloon catheter. A new method was
alsofound to improve the efficiency of foreign gene expres-sion in muscle by stitching medical
stitch containingplasmid DNA onto the quadricep muscle.
