大鼠子宫内膜离体培养4天后分组给药:对照组;丹那唑10-6,3×10-6,10-5mol/L;三烯高诺酮3×10-8,1O-7,3×1O-7mol/L;米非司酮10-6,10-5,10-4mol/L。于给药5天后取培养子宫内膜切片,镜下观察其形态变化,全自动图象分析仪细胞计数。结果表明,丹那唑抑制大鼠子宫内膜的生长,而三烯高诺酮和米非司酮未见明显抑制效应。人体子宫内膜离体培养4天后分组给药,1O-9mol/L雌二醇、10-7mol/L孕酮均能促进子宫内膜的生长。在给雌二醇或孕酮的同时加3×10-6mol/L丹那唑或10-7mol/L三烯高诺酮或1O-5mol/L米非司酮。结果表明,丹那唑、三烯高诺酮和米非司酮均能抑制子宫内膜的生长,而在雌二醇或孕酮缺乏的条件下,三烯高诺酮和米非司酮对子宫内膜的生长没有明显抑制作用。此结果表明三烯高诺酮和米非司酮对子宫内膜生长的抑制作用有赖于雌二醇或孕酮的存在。
The aim of the present study is to investigate the effect of gestrinone and mifepristone on the growth of rat and human endometrium in vitro. In vitro of rat endometrium was treated with l0-6, 3 × 10-6,1O-5mol / L danazol,3 × 10-8,1O-7,3 × 10-7mol / L gestrinone and l0-6 ,1O-5, 10-4mol / L mifepristone for 96 hours in separate groups. The morphologic features of endometrium were evaluated quantitatively by automatic image analysis system. The results showed that danazol inhibited the growth of endometrium, but no significant inhibition was detected in gestrinone and mifepristone groups. Endometrium of women in culture was treated with 10-9mol / L estradiol, 1O-7mol / L progesterone,3×x 10-6mol/ L danazol,1O-7mol/L gestrinone or l0-5mol / L mifepristone for 96 hours in separate groups.The results showed that estradiol or progesterone stimulated proliferation of endometrium, and these effects can be inhibited by gestrinone or mifepristone.
