摘要
目的探讨HIF-lα蛋白与IGF-1蛋白在人脑胶质瘤中的表达及两者之间的相关性。方法采用免疫组织化学SABC法检测HIF—lα、IGF-1表达蛋白在45例胶质瘤及10例正常脑组织中的表达。结果HIF-lα蛋白在10例正常脑组织中无表达,在45例胶质瘤中28例表达阳性,占62.2%,Ⅰ~Ⅱ级和Ⅲ~Ⅳ级间的表达有显著性差异(P<0.05)。IGF-1蛋白在10例正常脑组织中无表达,在45例胶质瘤中28例表达阳性,占62.2%,Ⅰ—Ⅱ级和Ⅲ—Ⅳ级间表达有显著性差异(P<0.01)。胶质瘤组织HIF-lα蛋白表达与IGF-1蛋白表达密切相关(γs=0.768,P<0.01)。结论HIF-lα蛋白异常表达与胶质瘤的发生有关,而IGF-1可能作为HIF-lα除缺氧以外的另一外诱导因素在HIF-lα促胶质瘤的发生过程中起重要作用。
Objective To explore the expression of HIF-1α and IGF-1 in human gliomas and the relationship between them. Methods Expression of HIF-1α and IGF-1 were assessed by immunohistochimistry staining in 45 cases of glioma and 10 cases of normal brain tissue. Results HIF-1α was expressed in 28 of 45 (62.2%) cases of glioma, and not expressed in the normal brain tissue.There were significant difference between low grade(I^IIgrade) and high grade(III^IVgrade) of glioma(P<0.05).IGF-1 was expressed in 28 of 45 (62.2%) cases of glioma, and not expressed in the normal brain tissue.There were significant difference between low grade(I^IIgrade) and high grade(III^IVgrade) of glioma(P<0.01). HIF-1α was significantly correlated with expression level of IGF-1(P<0.01,r_s=0.768). Conclusion The over expression of HIF-1α plays an important role in the apoptosis of glioma. IGF-1 probablely have revulsive use to HIF-1α in the apoptosis of glioma as a gene besides hypoxia.
