摘要
目的探索大肠癌及胆囊癌细胞株的最佳体外药敏实验条件。方法应用MTT比色测定法进行细胞生长曲线的测定,以了解两种细胞的对数生长期(最佳实验干预期)、最佳培养接种浓度等。结果分裂增殖快的大肠癌174细胞株可选用630nm波长进行吸光度测定,分裂增殖慢的胆囊癌细胞株450nm波长更优。结论大肠癌174细胞株的最佳细胞接种浓度为1.5~5.0×104/ml,对数生长期为接种后的3~5天;胆囊癌细胞株的最佳细胞接种浓度为1.4~2.6×104/ml,对数生长期为接种后的4~7天。
Objective To study the best experimental condition for chemosensitivity of human colorectal and cystic cancer cell line in vitro. Methods To measure the cellular growth curve using MTT colorimetric assay, in order to realize the logarithmic growth time of the two kinds of cell (the best experimental intervention time) and the prime density of cultivation. Results The best measure condition for the cell strain-174 of fast multiplication colorectal cancer was at a wavelengh of 630 nm, but the wavelengh of 450 nm is better measure condition for the cell strain of slow cultivation cystic cancer. Conclusion The former's best inoculation density is 1.5~5.0×10~4cells/ml, logarithmic growth time is the 3th^5th day after inoculation; but the catter's perfect density is 1.4~2.6×10~4cells/ml, logarithmic growth time is the 3th^7th day after inoculation.
