摘要 目的:建立组织原位核基质(NM)制备法。方法:用改进的硫酸铵沉淀法对食管癌组织冷冻切片进行NM制备。结果:经NM制备后切片的组织结构基本保持完好,细胞核淡染,呈丝网状,组蛋白H1染色阴性,FeulgenDNA染色弱阳性。结论:建立了可保持完好组织结构的原位NM制备法。 Objective:To establish a method of prepared tissue in situ nuclear matrix(NM).Method:The modified ammonium sulfate precipitating assay was used to prepare NM on frozen sections of esophageal carcinoma tissue. Results: After prepared the NM,the architecture of tissue was retained.NM was showed less intense staining material with reticular-like formation. Feulgen stained DNA was lightly positive staining and histone H1 was negative staining. Conclusion:The prepared method in situ NM with retained tissue architecture is established.
