摘要
IHW_2和IHW_3是亲溶酶体药物十二烷基吗啉(NDM)的类似物。由于分子中引入了亲水基团,所以是水溶性药物,较NDM更容易经胞饮作用进入富含溶酶体的白血病细胞,破坏溶酶体,从而导致细胞的死亡。IHW_2和IHW_3用量为85μg/1×10 ̄7和42μg/1×10 ̄7细胞时,,几乎能杀灭100%的白血病细胞,同时保留19.4±16.5%和7.9±12.7%的正常造血干细胞。IHW_3净化急性髓细胞白血病细胞的作用比IHW_2强约1倍。IHW_2和IHW_3有希望用于临床体外净化AML细胞。
HW_2 and IHW_3 were analogues of lysosomotropic agent N--dodecylmorpholine.Due to hydrophilic group they were soluble in water,and by pinocytosis easier enterinto lysosomes of leukemic cells,so as the lysosomes membrane rupured and the cells werekilled.Mean while 19.4±16.5% and 7.9±12.7% of normal stem cells were remained.The purging effect of IHW_3 was higher(1x) than IHW_2.So there will be possiblity touse IHW_2 and IHW_3 clinically for in vitro purging AML cells.
