摘要
用UV-Vis分光光度法研究了溴甲酚绿(BCG)与DNA基本的反应条件,BCG与DNA结合物的最大吸收波长是444nm,反应的最佳酸度为pH5.23;测定了反应时间、温度、离子强度、表面活性物质等因素对体系的影响;求出了BCG与DNA反应生成物的线性范围:(0.61~7.30)×10-7mol/L,表观摩尔吸光系数:1.29×105L·cm-1·mol-1,结合数:43,离解度:0.16,检出限为7.74×10-8mol/L,建立了DNA的反应体系。
The basic conditions for the interaction between bromocresol green(BCG)and DNA were studied by UV-Vis spectrophotometry.It was found that the maximum absorbance of BCG-DNA complex was atλ 444nm .The effects of interaction time and temperature were insignificant,but varied with the pH values.The optimum pH was5.23.The results also showed that the higher the ion strength,the lower the absorbance of the complex formed,and that the foreign ions and biological agents have certain interfering effects on the abˉsorbance.The linear range of the calibration curve for the BCG-DNA complex was(0.61-7.30)×10 -7 mol/L.The apparent molar absorptivity was1.29×10 5 L·cm -1 ·mol -1 with a detection limit of7.74×10 -8 mol/L.
