用^(125)I-HOSMcAb在骨移植成骨肉瘤组织的裸鼠体上的放射免疫显像 被引量:3

摘要 将人成骨肉肩细胞(HOS-8603)注射入裸鼠骨髓腔,建立了裸鼠骨荷人成骨肉瘤模型,并以 ̄(125)I标记抗人成骨肉启单克隆抗体( ̄(125)I-SIHOSMcAb-B_4),对该模型进行放射免疫显像,结果表明HoSMcAb-B_4对人成骨肉瘤组织的结合特异性较强,核素明显聚集于肿瘤处,而标记正常鼠IgG对荷人成骨肉瘤模型及标记抗体对荷人肝癌模型均未见肿瘤部位显像。这些结果提。示HoSMcAb-B_4抗体可用于成骨肉瘤的定位显像诊断及导向治疗的载体。 AbstractA xenograft of the tumour was established inthe bone of nude mice after injection of human os-teosarcoma cells ( HOS-8603)in the fembrs ofthese athymia animals。 The mice bearingxenografts showed localization of  ̄(125)I-OSMcAb-B_4 determined by both gamma-camera(ECT)imaging and organ distribution studies,Radiola。belled normal immunoglobulin( IgG)shOwed notumour localization as did radiolabelled HOSM-cAb-B_4 injected into nude mice bearing xenograftof human hepatocarcinoma。These results suggestthat the potential of the HOSMcAb-B_4 antibodyfor radioimmunodetection of osteosarcomas andpossibly as a carrier for therapeutic agents.
