摘要
为在短期内提高小麦品质,本研究构建了一个含有反义蜡质基因、HMWGS1Dy10基因和抗除草剂基因的重组质粒pWXAB,以优良小麦品种(冀麦24、白玉149、9411等)为材料,采用花粉管通道法进行转基因研究。对转基因后代进行除草剂筛选和SDSPAGE电泳。结果表明,除草剂基因及反义蜡质基因已经整合到小麦基因组中,从抗性水平而言,其转化频率为0.5%。
By pollen tube pathway,plasmid pWXAB containing anti-sense waxy genes,HMW-GS 1Dy10 gene and selective marker gene of bar were transferred into three wheat lines(JM-24、BY-149 and 9411). Half of the wheat seeds were used to examine waxy protein by SDS-PAGE,and the result indicated that anti-sense waxy genes had integrated into wheat genome. Other seeds were cultured and screened by herbicide,and then 12 transformed plants were obtained. For resistance to herbicide,the transformation rate was 0.5%.
