摘要
本文系统地分析了中国药典90版庆大霉素C组份的HPLC分析方法中流动相组成对庆大霉素C组份保留行为的影响,在SpherisorbC_(18)ODS柱上,最佳时流动相组成为0.05mol/L的庚烷磺酸钠:冰醋酸:甲醇:H_2O=4.7:1.3:73:21,此时各组份的相对保留时间约分别为0.13(衍生化试剂),0.27(C_1组份),0.65(C_1a组份),0.85(C2a组份)和1.00(C_2组份)。根据实验结果,对流动相中各组份在色谱分离过程中的作用进行了讨论。
The influence of mobile phase on the retention behavour of components C of Gentamicin was studied on the HPLC system employ-ed by Chinese Pharmacopia(1990).The optimum mobile phase was 0.05 mol/L sodium heptanesulphonate:glacial acetic acid:methanol:water=4.7:1.3:73:21 and the retention time of component pcaks with reiativ retent-ion times of about 0.13 (reagent),0.27(component C1),0.65(component C1a),0.85(component C2a)and 1.00(component C2)was about within 30 minutes on Spherisorb C18 ODS colunm(4 × 200mm).Also the mechanism of each component to the mobile phase in the chromatography was discussed based on experimental results.
