摘要
针对自主研制的抗真菌农用凯地菌素,以番茄为试材,通过比色法和聚丙烯酰胺凝胶电泳法系统地研究了4种处理(喷清水、唢凯地菌素、先喷凯地菌素再接种病菌和单独接种病菌)的番茄植株体内防御酶活性的变化、同工酶谱带的变化及对病程相关蛋白(PR)的诱导。试验表明:凯地菌素处理可使番茄植株POD、PAL、SOD、PPO的活性相对于对照均有不同程度的提高,酶活高峰集中表现在第4-10d,酶活性是对照的2-4倍;同1酶POD、PPO的电泳结果表明,喷药处理后可诱导番茄植株体内产生新的酶带,并且在第2-14d都有新增酶带,诱导抗性持久;同时,凯地菌素处理的番茄叶片细胞间隙中又诱导产生了两条新的PR蛋白谱带,经分析,该蛋白的分子量分别为26500D和16600D。
The effect of four treatments (a water spray, spray treatment with kaidimycin, spraying with kaidimycin followed by pathogen inoculation, and pathogen inoculation without kaidimycin) on induction of defense enzymes, isoenzymes, and pathogenensis-related proteins (PR) was systematically investigated in tomato plants using a specially formulated preparation of the agricultural fungicide kaidimycin, colorimetry, and polyacrylamide gel electrophoresis (PAGE). Kaidimycin treatment increased, to varying degrees, the activities of POD, PAL, SOD, and PPO compared to the control, with peak activity occurring from the 4th to the 10th day, and activities two to four times that of the control. In electrophoresis studies with isoenzymes of POD and PPO, kaidimycin induced new enzyme bands in tomato plants, with increases in the bands occurring from the 2nd to the 14th day after treatment, and indicating a long resistance induction. The production of two new PR protein bands was also induced in the intercellular spaces of tomato leaves treated with kaidimycin. The molecular weights of these new PR proteins were 26.5 and 16.6 kD.
