平肝熄风汤对大鼠脑出血后活化半胱氨酸天冬氨酸蛋白酶3表达的影响 被引量:1

摘要 目的:观察大鼠脑出血后血肿周围神经元活化半胱氨酸天冬氨酸蛋白酶3的表达情况及血肿变化,并评估平肝熄风汤的干预作用。方法:实验于2003-10在湘雅医院中西医结合研究所进行。用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型,免疫组化检测血肿周围活化半胱氨酸天冬氨酸蛋白酶3的表达,同时用量度尺测量大鼠血肿的最大直径。结果:Ⅶ胶原酶造模后模型组血肿周围于6h出现阳性表达细胞,1d阳性细胞计数达高峰,3d出现轻度下降,5d明显下降。与假手术组比较差异明显(P<0.01)。平肝熄风汤组与同时间点模型组比较:6h计数无明显差异,1,3,5d均存在明显差别(P<0.01)。造模后血肿于1d达到最大,3d有所变小,5d变小明显;治疗组在1d时与模型组比较血肿缩小明显(P<0.05),3,5d缩小更明显(P<0.01)。结论:大鼠脑出血后血肿周围神经元活化半胱氨酸天冬氨酸蛋白酶3明显活化,平肝熄风汤能抑制半胱氨酸天冬氨酸蛋白酶3的活化,阻止神经元的凋亡,同时促进血肿吸收。 AIM:To observe the expression of activated Caspase 3 in neuron around hematoma and the changes of hematoma in rats with intracerebral hemorrhage,and evaluate the intervention role of pinggan xifeng decoction. METHODS:The experiment was performed in the Institute of Combined Traditional Chinese and Western Medicine of Xiangya School of Medicine in October 2003.Collagenase Ⅶwas injected into rats with stereotaxic technique to build the ICH model.The immunohistochemical method was used to exam the expression of activated Caspase 3 in neuron around hematoma after intracerebral hemorrhage in rats. Meanwhile,the maximum diameter of hematoma at different time was measured with ruler. RESULTS:The positive cells appeared around the hematoma at 6 hours after operation, and peaked to the top at day 1,then decreased lightly at day 3 and it decreased distinctly at day 5.There was significant difference as compared with the sham operation group(P< 0.01).There was significant difference in the number of positive cells between the pinggan xifeng decoction group and the model group at days 1,3 and 5(P< 0.01), but not at 6 hours.The hematoma swelled to the biggest at day 1,deflated lightly at day 3, and became much smaller at day 5.The hematoma of treatment group deflated smaller than that of model group at 1 day(P< 0.05),and it deflated more obviously at 3 and 5 days(P< 0.01). CONCLUSION:The activated Caspase 3 increases obviously in the neuron around the hematoma after intracerebral hemorrhage;Pinggan xifeng decoction can markedly depress the expression of activated Caspase 3,prohibit neuronal apoptosis,and accelerate the absorbing of the hematoma.
