摘要
本文采用反相离子对高效液相色谱法测定了重瓣白曼陀罗、曼陀罗、紫花曼陀罗、无刺曼陀罗和毛曼陀罗在不同采收时期(开花初期、盛期、末期)花中的东莨菪碱和阿托品的含量。色谱条件为ZorbaxODS(4.6mm×250mm)柱,用含0.01mo1/L。辛烷磺酸的醋酸盐缓冲液(pH3.8)-乙腈(65∶35)为流动相,检测波长254nm。样品用甲醇作溶剂,超声提取40min。东莨菪碱和阿托品的平均回收率分别为98.28%和103.13%。不同采收时期的样品,以花开盛期为最高。
A reversed-phase ion-pair HPLC method was applied to isolate and determine the content of scopolamine and atropine in flower of Datura metel cv. cvata, D. stramonium var. stramonium.D. stramonium var. tatula. D. stramonium var.inermis and D. innoxia at different collective periods of time (initial, luxuriant, final). The separation was performed on a Zorbax ODS column (4. 6mm× 250mm) with a mobile phase of acetonitrile-0. 2mol/L acetate buffer (pH 3. 8)containing 0. 01mol/L octane sulfonic acid (35 : 65) . and detected at 254nm. The sample was treated by methanol in an ultrasonator for 40min. The average recoveries of scopolamine and atropine were 98. 28% and 103. 13% respectively.
