摘要
目的研究大鼠肝脏与DEN诱导肝癌高表达基因DD31完整序列的克隆。方法在以往采用DDPCR技术发现大鼠肝癌DD31EST片断的基础上,采用RACE-PCR的方法筛选雄性Wistar大鼠肝组织cDNA文库。结果首次得到DD31的全长表达的基因序列384bp,并实现GenBank的登录(GenBank登录号为AY653032)。经检索该基因定位于19号染色体19p12-p13区带。采用ExPASy服务器中的Translate和NCBI中的ORFfinder进行蛋白阅读框架(ORF)的识别,其中完整的ORF含有108bp,编码36个氨基酸。结论该基因在大鼠体内多组织均有表达,其中以肝组织、肝癌组织、脾组织和大脑组织中表达量较高。
Objective To study rat liver and the cloning of the complete sequence of DEN-induced hepatocarcinoma high-expression gene DD31.Methods On previous researches,RACE-PCR method was used to screen the cDNA series in the liver tissues of Wistar rats.Results The full-length expression gene sequence 384bp of DD31 was gained,and the GenBank login was carried out.This gene was located at 19p12-p13 area of the nineteenth chromosome.It was found that the complete ORF was 108bp,encoding 36 amino acids.Conclusions This gene happens in several tissuses,noticeably in liver tissue,liver cancerous tissue,spleen tissue and brain tissue.Heat-clearing and toxin-relieving therapy can down-regulate the gene expression in rat's liver cancerous tissue.
