血卟啉单甲醚-光动力疗法对Bcl-2的作用 被引量:9

摘要 体外研究表明,光动力疗法(PDT)主要是通过诱导细胞凋亡达到杀死细胞的目的。Bcl- 2 可以抑制凋亡的发生。为了研究血卟啉单甲醚(HMME)光动力诱导的 HeLa细胞凋亡对 Bcl- 2 的作用,收集照射(15 mW/cm2,5.4 J/cm2)后0、2、4、6 h的细胞,用半胱氨酸天冬氨酸特异性蛋白酶(caspase- 3)的活性表征细胞凋亡的发生;通过western杂交检测PDT处理后细胞内 Bcl- 2 含量的变化。研究发现,PDT处理后与0 h相比,2、4、6 h后caspase 3的活性有增高的趋势,但Bcl- 2的含量没有明显的变化。 Photodynamic therapy (PDT) can induce cell apoptosis. Bcl-2 family is the key regulators in apoptosis. In this study, we assessed the effects of hematoporphyrin monom-ethyl ether (HMME) mediated PDT on Bcl-2, Followed irradiation (15 mW/cm2, 5.4 J/cm2), monolayer HeLa cell were harvested at 2, 4 and 6 hours respectively. The apoptosis was confirmed by the detection of the activity of caspase-3. The content of Bcl-2 was determined by Western blotting. HMME-PDT can induce HeLa cells apoptosis as the activity of casase-3 significantly increase, but the content of Bcl-2 of treated cells are not significantly different from that of control cells.
