摘要 提出了从裸子植物侧柏中分离纯化核糖体失活蛋白的制备和检测方法.通过盐溶液抽提、硫酸铵分级沉淀、阴离子交换层析、阳离子交换层析等步骤制备了该蛋白.该蛋白的检测方法是把这种蛋白作用于大鼠 80S肝核糖体后, 不经过酸性苯胺处理,直接进行聚丙烯酰胺的尿素变性胶电泳.在电泳图上得到 400 bp左右的特征性核酸条带.分析结果表明,该蛋白为一种新的核糖酶型的核糖体失活蛋白,阐明了侧柏核糖体失活蛋白失活核糖体的分子机制. The preparation and detection methods of isolating and purifying a ribosome-inactivating protein (RIP) from the seeds of arborvitae were presented. RIP was prepared by extraction with salt solution, ammonium sulfate fractional precipitation, anion exchange chromatography and cation exchange chromatography. 80S ribosomes RNA (rRNA) from murine liver were treated with RIP directly without acid aniline treatment. The polyacrylamide gel chromatography figure showed a character nucleic acid fragment approximately 400 bp. The analytic results show that the protein is a new RIP with specific ribonuclease activity. The functional mechanism of this arborvitae RIP at the molecular level is illustrated.
