摘要
目的揭示吲哚美辛抑制细胞增殖及诱导细胞凋亡的分子机理,探讨其抗结肠肿瘤的作用机制。方法不同浓度的吲哚美辛作用于结肠癌细胞株HCT116细胞24h,采用Western蛋白印迹技术检测CDK2、CDK4、Bcl-2、Bax及p21WAE1/CIP1蛋白表达。结果吲哚美辛能降低CDK2、CDK4及Bcl-2的表达,同时上调p21WAE1/CIP1蛋白的表达且呈剂量依赖方式,而对促凋亡蛋白bax的表达无影响。结论吲哚美辛可通过降低CDK2、CDK4、Bcl-2蛋白,上调p21WAF1/CIP1蛋白的表达来抑制细胞增殖和诱导细胞凋亡,从而实现其抗肿瘤作用。
To explore the molecular mechanism of indomethacin on proliferation and apoptosis in the HCT116 cell line and provide experimental evidences that IN was used for colon cancer in clinic. HCT116 were treated with different concentrations of IN for 24 h and then CDK2, CDK4, Bcl-2, Bax and p21WAF1/CIP1 proteins were detected by Western blotting. The expression of CDK2, CDK4 protein were down-regulated and the expression of p21WAF1/CIP1 protein were up-regulated in HCT116 cells treated with different concentrations of IN ,in dose-depentent increasing from 100 μmol/L to 600 μmol/L. Both of expression Bcl-2 protein and the ratio of Bcl-2/Bax were down-regulated, in parallel to the increase of IN dose, but the expression of Bax did not change. [Conclusions] Our results indicate that molecular mechanisms of IN inhibiting proliferation and inducing apoptosis are attributed to down-regulating the expression of CDK2, CDK4, Bcl-2 proteins and up-regulating p21WAF1/CIPI protein.
