胎脑中神经生长因子(NGF)的提取及生物活性鉴定的研究 被引量:4

摘要 通过匀浆、离心、除脂、超滤及柱层析等过程,从中小月份引产的人胚胎脑中提取出较纯的NGF。经SDS-聚丙烯酰胺凝胶平板电泳发现其分子量为16000和36000的二个组分,大于小鼠NGF14000的分子量;另外,其氨基酸组成与蛇及小鼠NGF略有不同。本提取液能维持并促进鸡胚脊髓和背根神经节神经细胞在体外的存活和生长,具有良好的生物学活性及稳定性;毒性试验证实无刺激性及毒副作用。 The study is caried out by using plasma through aseries of processes as centrifugation, freeze-defatting and sephadex G-100 filtration chromatography. By means of SDS polyacry slab gel eletrophoresis.the purified NGF from the brain of the human embryo is proved to be of two components. Their molecular weights are 36000, 16000 nespectively. So it can be infered that the chains and its amino acid composition is slightly different from that of NGF in the snake and mouse . which may be the result of species differences. The extract of the NGF keeps the nerve cells of the chick embryo's spinal cord and dorsal root ganglions living and growing in vitro. Bio active assay shows that when the NGF extract concentration reaches 200μg/ml, the nerve tissue cul ture will get to the best state.
