缺血性肾损伤中程序化细胞死亡形态及分子特征 被引量:4

Morphologic and molecular evidenceof Apoptosis during the reperfusionphase after brief periods of renalischerma

摘要 本文观察了缺血性肾损伤中肾小管上皮细胞死亡的另一种形式—Apoptosis。通过钳夹肾蒂造成小鼠缺血冉灌注肾损伤模型,结果发现,缺血5min再灌注12~24h肾小管上皮细胞并不出现坏死,但可发生Apoptosis现象。随着缺血时间的延长,缺血30min再灌注6~24h,或缺血45min再灌注6~18h,肾小管和间质出现Apoptcsis小体,肾脏DNA电泳像呈“梯形结构”。梯形结构的出现持续24h后消失。结果提示,缺血再灌注肾损伤中存在Apoptosis机现的参与,Apoptosis小体和梯形结构可作为缺血性肾损伤的早期观察指标。 A multiparanietric analysis to demonstratethat even breif poriod of ischemia can initiateextensive lase in a rat kidney through the processof Apoptosis during early reperfusion.Microscopic exaniination of mouse renal tissuessubJecteil to a 5-, 30-, or 45- minute periodcomplete ischeinia showed the presence ofapoptotic bodies .both within and occasionallybetween renal tubular, aplwaring as early as 6houro after reperfusion and increasulg innumbers at 12 hours. Furtherniore, DNAextracted forrn such reperfusion renal tissuedemonstrated the appearance of a distinct“ladder pattern” of DNA fragments afterelectrophoresis in agnase gels. It was sugsestedthat renal reperfusion injury after ischemia couldinitiate a form of cell death-Apoptosis that isdrastically different from eellular necrosisinduced by prolonged severe ischemia.
