摘要
目的 :探讨 8种L 支链氨基酸合剂对急性肾缺血 再灌注损伤的保护作用及作用机制。方法 :夹闭大鼠双侧肾蒂 4 5min ,再灌注 2 4h制成肾缺血再灌注损伤动物模型 ,将 2 4只SD大鼠随机分为假手术组 (A组 )、对照组 (B组 )、氨基酸治疗组 (C组 ) ,通过检测尿量、血尿素氮 (BUN)、血肌酐 (Cr) ,肾组织苏木精 伊红染色及用免疫组织化学进行bcl 2和HSP 70表达检测。结果 :3组肾组织中bcl 2和HSP 70均有表达 ,但免疫反应强度不同。与A组相比 ,B组尿量增加 ,血BUN、Cr升高 ,肾小管上皮细胞呈现缺血性改变 ,HSP 70表达明显增强 ,两组间差异有显著性意义 ,而bcl 2表达差异无显著性意义。与B组相比 ,C组尿量明显增加 ,血BUN、Cr明显下降 ,且缺血损伤程度有减轻 ,bcl 2表达明显增强 ,两组间差异有显著性意义 ,而HSP 70表达差异无显著性意义。结论 :该氨基酸合剂对大鼠急性肾缺血再灌注损伤有明显保护作用 ,其作用机制可能与它影响bcl 2表达有关。
Purpose:To explore the protective effects and the beneficial mechanism induction after amino acids preconditioning on acute renal ischemic reperfusion injury(ARI) in rats. Methods:A model of bilateral post ischemic renal injury by clamping renal pedicles for 45 minutes was set up. 24 SD rats were divided randomly into 3 groups:sham operated group (n=8);ischemic control group (n=8);amino acids preconditioning group(n=8). We examined unine volumn, renal function. Histology and the expression bcl 2 and HSP 70 by immunohistochemical method. Results:HSP 70 and bcl 2 were observed in all groups. As Compared with those of Sham operated control group,rate of urine flow, serum creatinine,BUN values and HSP 70 expression increased significantly (P< 0.05 ) in the ischemic group. But, bcl 2 expression had no significant effect on the two groups (P< 0.05 ). As compared with those of the ischemic control group, urine volumn and bcl 2 expression increase remarkably in the amino acids group, and serum creatinine and BUN values reduced (P< 0.05 ), but HSP 70 express had no significant effect on the two groups.Conclusions:This mixture of 8 L amino acids provided remarkably protection against ARI in rats,and the protective effect was associated with up regulation of bcl 2.
