双特异抗体体外模拟特异B细胞抗原递呈作用

摘要 用化学偶联的双特异性抗体复合物-αTT-αIgM通过体外实验。观察其抗原递呈能力。结果显示:αTT-αIgM存在时,可使非特异性B细胞的抗原递呈效应显著提高,所需抗原浓度较不加αTT-αIgM的对照组低1000倍左右,与抗原特异性B细胞的抗原递呈效应相似。抑制试验显示:羊抗鼠IgM,鼠IgM能抑制αTT-αIgM对非特异性B细胞抗原递呈作用的增强效应,表明B细胞膜IgM在抗原递呈中起关键性作用。本研究结果表明:αTT-αIgM的存在确能模拟抗原特异性B细胞的高效抗原递呈功能。 By two independent assay(T cell proliferation &IL-2 secretion)the effect of bispecific antibody complexby chemical conjugated-aTT-aIgM on presentation of antigen was investigated,In the presence of aTT-aIgM.the abilityto present antigen of non-specific B cell was significantly enhanced and the antigen(TT) was presented at concentration10 ̄3-fold lower than that by non-specific B cell without aTT-aIgM or by M.but just the same as the antigen-specific Bcell.We also observed that the enhanced presentation was significantly inhibited by mouse IgM or anti-mouse-IgM. Theseresults indicated that high efficiency presentation of antigen-specific B cell can be mimicked by aTT-aIgM.
